AB - Background: Quercus infectoria is one of the most important medicinal plants in the Zagros mountains, which from the ancient time has been taken into consideration as a known medicinal plant. Studies have shown, this species contains a wide range of medicinal properties. In this research comprehensive introduction of this medicinal tree, places of distribution and manners of correct diagnosis have been proposed. Materials and Methods: Specimens were collected from natural habitats in different regions of Zagros. After identification, normal leaves were used to extract DNA and SEM studies. To maintain the quality of DNA, fresh leaves were dried on silica gel. After extraction of DNA, purification of PCR product and sequencing, nucleotide sequences were edited using Sequencher software. Results: In oak galls, fruits matured in one year, whereas in other species fruits matured in biennial. In this species cuppule is sessile or semisessile. Margin of leaves is complete or lobate. SEM images showed that the leaves are glabrous or posses sparse and scattered trichomes. Nucleotide sequence of DNA is about 680 bp. Conclusion: The introduction of oak galls has been done to identify and correct exploitation of this valuable medicinal plant, and with determination of its DNA barcoding, the rate of purity or fraud of the samples in the market is evaluated.
