Background: Gastric cancer is one of the most common and lethal cancers worldwide. Due to its late-stage diagnosis, finding new therapeutic approaches is of significant importance. In this regard, the present study aimed to evaluate the cytotoxic effects of green-synthesized selenium nanoparticles using rosemary plant extract (Salvia rosmarinus) on gastric cancer cells (MKN-45).
Materials and Methods: In this experimental-laboratory study, selenium nanoparticles were synthesized via a green synthesis method using Salvia rosmarinus extract. The MKN-45 gastric cancer cells were treated with specified concentrations of the nanoparticles. Cell viability and proliferation were assessed using the MTT assay. Following treatment and subsequent incubation, the cytotoxic effects of the nanoparticles were evaluated by measuring the optical absorbance of the formazan solution. Cell viability percentage and IC50 were calculated and analyzed using analysis of variance statistical tests.
Results: The results showed that selenium nanoparticles at concentrations of 31.25, 62.5, 125, 250, and 500 µg/mL significantly reduced the viability of gastric cancer cells, and cytotoxicity increased with higher concentrations (p < 0.001). The IC50 analysis indicated that increased SeNP concentration led to decreased cancer cell survival. Physicochemical analyses, including Fourier Transform Infrared, Scanning Electron Microscopy, and Energy Dispersive X-ray, confirmed the successful synthesis of selenium nanoparticles with appropriate structural characteristics.
Conclusion: Results of this study demonstrated that green-synthesized selenium nanoparticles exhibit significant cytotoxic effects on gastric cancer cells (MKN-45) and may serve as a promising candidate for the development of targeted therapies against gastric cancer.
