Background: The present study was designed and conducted to evaluate the protoscoccidial effects of ethanolic extract of Nectascordeum koelzi root on hydatid cyst protoscoleces in both in vitro and ex vivo conditions.
Materials and Methods: Protoscoleces (1×10³/ml) were incubated with different concentrations of plant extract (2.5, 5, 10, and 20 mg/ml) for different times (5, 10, 20, 30, and 60 minutes). Then, the viability of protoscoleces was determined using 0.1% eosin staining test. The effect of the extract on caspase-3-like enzyme activity was also investigated using a colorimetric method.
Results:  By in vitro experiments the plant extract at concentrations of 10 and 20 mg/ml caused 100% lethality of protoscoleces after only 10 and 30 minutes of exposure. On the other hand, the ex vivo results showed that the extract at similar concentrations was able to induce complete lethality (100%) of protoscoleces after 20 and 60 minutes of exposure, respectively (p<0.001). The extract increased caspase-3 activity in protoscoleces in a concentration-dependent manner.
Conclusion: Given its rapid and significant effect on hydatid cyst protoscoleces, this extract can be considered as a promising alternative to conventional chemical scolicides during hydatid surgeries.
